Chronic systemic pesticide exposure reproduces features of Parkinson's disease. Nature Neuroscience 3, 1301-1306

| The cause of Parkinson's disease is unknown, but an association with pesticides and other environmental toxins is suggested, and biochemical studies implicate a systemic defect in mitochondrial complex I. The chronic systemic inhibition of complex I by the lipophilic pesticide rotenone, is shown to cause highly selective nigrostriatal dopaminergic degeneration, associated with hypokinesia and rigidity. Exposure to naturally occurring compounds or synthetic pesticides that are potent inhibitors of complex I, combined with genetic differences in complex I function, may underlie most cases of typical idiopathic Parkinson's disease.
